چکیده
PURPOSE To investigate the in vivo expression of toll-like receptor 4 (TLR4) and its associated lipopolysaccharide (LPS) receptor complex in the human eye. METHODS Normal human ocular tissues were evaluated for in vivo TLR4, MD-2, and CD14 mRNA and protein expression by RT-PCR and immunohistochemistry, respectively. The distribution patterns and phenotypes of the cells expressing these proteins were further characterized by confocal microscopy and double-label immunofluorescence studies. RESULTS Normal human uvea, retina, sclera, and conjunctiva constitutively expressed TLR4, MD-2, and CD14 mRNA. The protein expression of these molecules was restricted, however, to resident antigen-presenting cells (APCs) in the normal human uvea, consisting mainly of HLA-DR(+) dendritic cells (DCs). These APCs endowed with the complete LPS receptor complex appeared to be strategically positioned in perivascular and subepithelial locations for surveying blood-borne or intraocular LPS. In contrast, other cell types of the normal human cornea, conjunctiva, retina, and sclera did not express TLR4/MD-2 protein in vivo as detectable by immunohistochemistry. CONCLUSIONS The present study demonstrates for the first time that resident APCs in the normal human uvea express TLR4 and its associated LPS receptor complex. This has significant implications for the understanding of normal ocular immunity as well as unraveling the potential role of Gram-negative bacteria in the pathogenesis of acute anterior uveitis (AAU).

P-33: Expression of Toll-Like Receptor 2-3 Genes in Human Sertoli Cells
Background: Toll-like receptors (TLRs) constitute a major part of innate immunity, which can distinguish pathogen associate molecular pattern. Sertoli cells create a special immunological niche that protects somniferous tubules from auto antigens and pathogens. These cells are the only somatic cells in somniferous that protect testis cells against pathogens. The purpose of this study was to eva...
متن کاملLipopolysaccharide Induced Activation of Toll Like Receptor 4 in Isolated Rat Heart Suggests a Local Immune Response in Myocardium
Background: Myocardial dysfunction is one of the major complications in patients with sepsis where there is a relationship between the blood level of cytokines and the onset of myocardial depression. In many cases of sepsis, the presence of Lipopolysaccharide (LPS) has been established. LPS Binding Protein (LBP) bound endotoxin is recognized by CD14/toll-like receptor-4 (TLR4) complexes in inna...
متن کاملSilymarin inhibits Toll-like receptor 8 gene expression and apoptosis in Ramos cancer cell line
Objective: Silymarin is a herbal extract containing flavonolignans, and it has inhibitory effects against the growth of different cancer cell lines by inducing apoptosis. Toll-like receptors are suggested as a novel and attractive target to treat cancer. The current study aimed at examining the mechanism of silymarin-induced apoptosis in Ramos cells and investigating its effect...
متن کاملI-23: Reproduction and Toll Like Receptors(TLRs
Female and male reproductive tracts are of interest sites to study of immune system because they encounter specific infections such as those are sexually transmitted. Furthermore, female reproductive tract is in close contact with allogenic sperms and transmitted microorganisms during intercourse and semi allogenic fetus during pregnancy. In mammals, there are two types of immune responses, the...

Background: During pregnancy, the maternal immune system is in close contact with cells and tissue from the semiallogenic fetus .The human leucocyte antigen (HLA) class Ib molecules, HLA-E, -F and -G, are expressed at the materno-fetal interface. Because of the apparent immunoregulatory functions of these proteins, they may be involved in successful acceptance of the semi-allogenic fetus during...
